Nicotinamide adenine dinucleotide (NAD+) is a dinucleotide coenzyme found in all living cells, playing a central role in redox reactions and energy metabolism. It exists in oxidized (NAD+) and reduced (NADH) forms and serves as a substrate for multiple enzyme classes including sirtuins (SIRTs), poly(ADP-ribose) polymerases (PARPs), and CD38.
Preclinical research has investigated NAD+ in the context of mitochondrial biogenesis, sirtuin-mediated deacetylation, DNA damage repair via PARP activation, and circadian rhythm regulation. Studies have also examined its role in cellular senescence, neuronal energy metabolism, and age-related decline in NAD+ biosynthesis pathways including the salvage pathway via NAMPT.
